Lieshman Staining Procedure

Lieshman Staining का प्रयोग ब्लड स्मीयर के माइक्रोस्कोपी के लिए किया जाता है जो मलेरिया पैरासाइट, DLC और एनीमिया को डायग्नास करने मे मदद करती है।

Used for microscopy of peripheral blood smear or general blood picture for Diagnosis of Malaria parasite, Differential leukocyte count and other anemic Diseases.

                                                                                                            Lieshman Staining –

सबसे पहले एक Slide पर Blood Smear  बनाकर सुखा लेते है और जब Slide सूख जाता है तब उसे Stain करते है।

Preparation of Lieshman stain solution –

Lieshman Stain Solution

  Requirements (आवश्यक सामग्री) – 

1) Lieshman Powder – 0.6 gm

2) Methanol – 400 ml

3) Dry container

4) Graduate cylinder

5) water bath

       

Procedure (Stain Solution बनाने की विधि) –

1) सबसे पहले एक dry container में 0.6 gm Lieshman Powder लेते है।

2) अब Graduate cylinder की सहायता से 400 ml Methanol नाप लेते है।

3) अब वाटर बाथ जिसका तापमान 37 डिग्री रखते हैं में एक dry container लेते है और उसमें 0.6 gm Lieshman Powder और 400 ml Methanol मिला देते है और तब तक गर्म करते है जब तक Lieshman Powder Methanol में अच्छे से घुल न जाए।

4) Solution तैयार होने के बाद इसे Cool और Dark place में रखते है।

5) Solution का प्रयोग करने से पहले इसे फिल्टर पेपर से छान लिया जाता है।

नोट - इसका प्रयोग अधिकतम 3 महीने तक किया जा सकता है।

Preparation of Buffer Solution –

Buffer Solution

Requirements (आवश्यक सामग्री) – 

1) Distilled Water – 1 liter

2) Ph Tablet – 1 tab

3) Clean Dry Container               

Procedure (Buffer Solution बनाने की विधि) –

1) सबसे पहले एक dry container लें और उसमें 1लीटर Distilled Water भरें।                                    

2) अब उसमें 1 Ph Tablet मिला दें और Tablet को पानी में घुलने के लिए छोड़ दें।

3) इस प्रकार बफर साल्यूशन तैयार कर लिया जाता है।

Note – Lieshman stain solution and buffer solution are also commercially available.

Stained Slide Under Microscope

Requirements (आवश्यक सामग्री) –

 1) Dry slide With blood smear

 2) Slide Stand

 3) Lieshman stain

 4) Buffer solution

 5) Tap water or Dropping Bottles

 6) Cotton wool

Procedure Of Staining –

1) सबसे पहले smeared Slide को 15 sec. -1min. के लिए Lieshman stain की 7-8 बूँदो से cover कर देते है।

2) अब 10-15 min. तक Buffer solution की दोगुनी मात्रा slide पर डालते है और Mouth से सावधानीपूर्वक फूँककर buffer और Stain को mix करते हैं।

3) समय पूरा होने पर slide को tap water  से या drop bottle की सहायता से धुल लेते हैं और slide धुलने के समय slide के पीछे वाले भाग को Cotton wool से पोछते रहते है ताकि slide अच्छे से साफ हो जाए।

4) अब slide को सुखा लेते है और फिर यह microscopic examination के लिए तैयार हो जाता है।
